Carcinoembryonic antigen (CEA) is an acidic glycoprotein with a human embryonic antigenic determinant with a molecular weight of 150-200 kD. The CEA-encoding gene is located on chromosome 19. CEA is a large family of cell surface glycoproteins and is part of the immunoglobulin gene superfamily. Fetal and adult digestive tract produce a small amount of CEA, normal CEA is secreted into the gastrointestinal tract, while cancer cells that lose polarity secrete CEA into the blood and lymph, so the CEA in the blood increases.

When the concentration of CEA in the sample is high, the remaining antibody binding sites are small, so that 125I-CEA bound to the antibody is less. The antigen-antibody conjugate was separated using a separating agent, and the radioactivity in the conjugate was determined. It was found that the binding amount of 125I-CEA was a function of the concentration of CEA in the sample. After plotting the data of the standard points into a function curve, you can find the CEA concentration value in the specimen.
